Crucian carp graph: distinguish treatment protection from proposed host mediation #1352

Closed

Description

@realmarcin

Source-based self-adversarial review of CommunityMech:000293, Crucian_Carp_Gut_Disease_Resistance_SynCom.yaml, against complete primary main body PMID:41280275 / PMC12637255.

The three nodes use COMMENSALISM, MUTUALISM and COMPETITION without microbial partner fitness evidence. The barrier node promotes transcript measurements into protein/functional barrier confirmation; both output arrows overstate selective mediation. The IgA phrase is a KEGG pathway annotation, not an IgA production assay.

Acceptance: retain three nodes/two arrows and canonical genus participants. Preserve the 28-day SynCom intervention, enzyme activities, qPCR-validated transcripts, challenge survival (60% vs 32.5%) and calibrated qPCR pathogen-load reductions. Distinguish these from FMT survival (66.7% vs 36.7%). Qualify both mechanistic arrows, keep elevated DAO as an ambiguous injury/turnover marker, and avoid reciprocal microbial fitness claims. Record lack of mediator-specific blockade/rescue, direct barrier permeability/protein validation and exact in-host partner mechanisms. Add history, focused regressions and regenerated page.

Primary Methods, Results, Discussion, Conclusion and displayed captions read; no figure images or supplementary files inspected. Preliminary single/pair/triple feeding results are reported in main text, not independently checked in S11. Closed #529 concerns cultivation provenance, not these graph claims.

Dedup: 687 all-state titles/bodies and hidden/ignored-inclusive local files searched; no exact graph duplicate in that scope. Comments not exhaustive. Source-based self-review, not independent approval.
